目的:通过细胞外过氧化氢(H_2O_2)的刺激建立单个人肺癌SPC-A-1细胞的氧化压力模型。创新点:氧自由基(ROS)涉及多种生物现象,包括有益和有害两个方面。ROS的定量检测和反应网络的评估结果令人期待。但ROS半衰期很短且反应过程很快,因此,我们通过多种手段克服了检测和评估的困难。方法:利用改进的微流控和成像技术测定ROS水平,构建氧反应网络。通过调控线粒体胞浆Ca2+水平、线粒体Ca2+摄取、细胞内ROS自扩增以及内在凋亡途径,确定线粒体在外源氧化压力模式中扮演的角色。结论:研究结果表明1 mmol/L H_2O_2引起细胞O_2·-水平的快速增加,从而导致细胞氧化能力增加和还原能力降低。此外,研究还证实了内质网中储存的Ca2+是H_2O_2诱导的线粒体Ca2+爆发的主要来源。外源氧化压力反应涉及细胞器间Ca2+信号的传递、ROS自身扩增、线粒体功能紊乱和半胱天冬酶依赖性凋亡途径。线粒体在外源性氧化应激影响细胞命运方面发挥着关键作用。 相似文献

Bamboo was a popular material substituting for wood,especially for one-off commodity in China. In order to recover energy and materials from waste bamboo,the basic characteristics of bamboo pyrolysis were studied by a thermogravimetric analyzer. It implied that the reaction began at 190~210 °C,and the percentage of solid product deceased from about 25% to 17% when temperature ranged from 400 °C to 700 °C. A lab-scale fluidized-bed furnace was setup to research the detailed properties of gaseous,liquid and solid products respectively. When temperature increased from 400 °C to 700 °C,the mass percent of solid product decreased from 27% to 17% approximately,while that of syngas rose up from 19% to 35%. When temperature was about 500°C,the percentage of tar reached the top,about 31%. The mass balance of these experiments was about 93%~95%. It indicated that three reactions involved in the process: pyrolysis of exterior bamboo,pyrolysis of interior bamboo and secondary pyrolysis of heavy tar. 相似文献

Barry C.Keenan has been acknowledged as an influential American Sinologist since his doctoral dissertation entitled The Dewey Experiment in China:Educational Reform and Political Power in the Early Republic was published by Harvard University Press in 1977.His deft approach to the sociological history of Chinese education was further exemplified in his second monograph,Imperial China's Last Classical Academies:Social Change in the Lower Yangzi,1864-1911,published by the University of California Press in 1994.This academic background suggests that Keenan had been actively involved in the new cultural history movements that sprung up in the 1970s. 相似文献

Seaweed has been used in traditional cosmetics and as a herbal medicine in treatments for cough,boils,goiters,stomach ailments,and urinary diseases,and for reducing the incidence of tumors,ulcers,and headaches.Despite the fact that seaweeds are frequently used in the practice of human health,little is known about the role of seaweed in the context of inflammation.This study aimed to investigate the influence of Jeju endemic seaweed on a mouse macrophage cell line(RAW 264.7) under the stimulation of lipopolysaccharide(LPS).Ethyl acetate extracts obtained from 14 different kinds of Jeju seaweeds were screened for inhibitory effects on pro-inflammatory mediators.Our results revealed that extracts from five seaweeds,Laurencia okamurae,Grateloupia elliptica,Sargassum thun-bergii,Gloiopeltis furcata,and Hizikia fusiformis,were potent inhibitors of the production of pro-inflammatory mediators such as nitric oxide(NO),prostaglandin E2(PGE2),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).Based on these results,the anti-inflammatory effects and low cell toxicity of these seaweed extracts suggest potential thera-peutic applications in the regulation of the inflammatory response. 相似文献

Studies of synergetic irradiation effects and liquid lead-bismuth eutectic (LBE) corrosion/embrittlement effects on ferritic/martensitic (F/M) steels are of great importance for developing high power spallation neutron targets (〉1 MW) such as the European Spallation Source (ESS) and Accelerator Driven System (ADS) facilities that can be used for transmuting long-lifetime radioactive wastes. Liquid LBE (45Pb-55Bi, in terms of mass fraction) has been selected as the candidate target material in high power spallation neutron targets due to its favourable thermal, physical & chemical properties, and to its high spallation neutron yield. 9Cr F/M steels such as T91 (9CrlMoVNb, in terms of mass fraction) have been chosen as the structural material for the targets due to their good mechanical properties and good resistance to irradiation induced swelling in fission neutron irradiation environments. For developing high power spallation neutron targets, behaviors of F/M steels in spallation neutron target irradiation environments and LBE corrosion/embrittlement effects have been extensively studied. However, many open questions have not been answered. The aim of this paper is to describe the present research situation on this topic. The obtained experimental data about LBE embrittlement effects on F/M steels is summarized and the influence of different parameters involved is analyzed to understand the influence effect on LBE embrittlement effect of F/M s. 相似文献

Objective:High body mass index (BMI) is considered as the most important risk factor for elevated serum alanine aminotransferase (ALT) concentration. This study examined an array of factors, including waist circumference (WC) and folate deficiency, which may mediate the association of BMI with serum ALT concentration in Chinese hypertensive adults without known hepatic diseases. Methods:A multicenter, cross-sectional study was carried out. A total of 378 patients with mild or moderate hypertension and without known hepatic diseases were recruited from five hospitals in Harbin, Shanghai, Beijing, Xi’an, and Nanjing. Results:Of the 360 hypertensive patients with complete data in our final analysis, 13.6% had high ALT concentrations (>40 IU/L). Factors including BMI, WC, triglyceride level, and folate concentration were associated with ALT concentration in univariate analysis. Consistently higher prevalence rates of elevated ALT were observed in subjects with lower folate concentrations (≥12 vs. <12 nmol/L, 9.9% vs. 17.8%, P=0.03), with higher BMI (≥28 vs. <28 kg/m2, 21.5% vs. 11.4%, P=0.02) or higher WC (≥90 vs. <90 cm, 18.5% vs. 10.0%, P=0.02). However, in multivariate analysis, the association between BMI and ALT concentration disappeared (P=0.802 in males and 0.369 in females), while WC in females (P<0.001) and folate concentration (P=0.036 in males and 0.044 in females) remained as significant predictors for ALT concentration. Conclusions:This multicenter study demonstrated that WC and low folate concentration were important factors underlying the association between BMI and ALT concentrations in Chinese hypertensive adults without known hepatic diseases. 相似文献
